Since the introduction of the term proteomics (1), different methodologies have been developed that allow global analyses of gene expression at the protein level. The original papers mainly described the analysis of differential gene expression using two-dimensional polyacrylamide gel electrophoresis (2-D-PAGE). Nowadays, beside 2-D-PAGE, approaches involving differential isotopic labeling (2,3) or multidimensional chromatography (4) have been introduced. Although these new methods certainly have significant advantages, particularly in terms of quantitation, they should rather be regarded as being additional and, so far, they do not completely replace differential 2-D-PAGE analysis, as stated in reviews by several authors (5,6). Also, in other research areas related to proteomics, gel electrophoretic methods are being used. A striking example is the study of the interactome.
