Opioid receptors belong to the family of the CTP-binding proteins coupled receptors. In in vitro systems much work has been done to characterize the subtypes of G proteins regulated by opioid receptors. However, to achieve this goal in vivo is a complicated issue. Pioneering studies involving pertussis toxin demonstrated the regulation of G(i)/G(o) to produce supraspinal opioid analgesia. In characterizing those G proteins regulated by mu and delta opioid receptors to induce supraspinal analgesia in the mouse, our group has explored the effectiveness of in vivo administration of antisera raised against different Gα subunits and antisense oligodeoxynucleotides to their mRNAs.
