LIMBAL STEM CELL DEFICIENCY MODEL IN RABBITS FOR EXPERIMENTAL CORNEAL SURFACE REGENERATION

In this study, we present a modified technique for creation of limbal stem cell deficiency in rabbits. Twenty percent isopropanol was used to completely remove rabbit corneal epithelium, and then superficial lamellar limbectomy (developed by other investigators) was performed to excise most or all limbal epithelial stem cells. Rabbit eyes were examined at least once every month for 3 months with slit lamp, fluorescein staining, impression cytology (IC), and western blot analysis after LSCD creation. Slit lamp exams indicated that the severity of corneal vascularization and conjunctivalization did not stabilize until 2–3 months after LSCD creation. IC showed many macrophages on the corneal surface at one month, reduced but still significant macrophage infiltration at two months, and very few macrophages with many fibroblasts on the central corneal surface at 3 months.
