Cell suspension culture of Viola diffusa Ging.

TL;DRAbstract

The conditions of cell suspension culture and growth kinetics of Viola diffusa Ging were studied. The results showed that the fittest inoculating quantity of Viola diffusa Ging in cell suspension culture was 2.0~3.5 g (50 mL). The fittest culture media were MS+2.0 mg/L ?2,4 D+1.0 mg/L ZT+300 mg/L LH+200/L sucrose (or glucose), with pH 5.8. The cell number development of Viola diffusa Ging during a culture course was similar to a “S” curve with apparent logarithmic phase. Contrast to the cell number, fresh weight ( m FW ) and dry weight ( m DW ) had no apparent logarithmic phase, but a gradually rising tendency appeared. There was also a relationship between cell multiplication and pH during the suspension cell culture of Viola diffusa Ging. Fig 3, Tab 2, Ref 12
