Effects of hydrogen peroxide on neuronal firing in cultured rat celiac neurons

Elevated reactive oxygen species (ROS) and altered patterns of firing associated with reduced potassium currents were found in celiac ganglion neurons from hypertensive rats. In this study, we investigated whether these changes could be mimicked by application of H 2 O 2 . Using whole‐cell patch clamp, dissociated celiac neurons from 3‐week old SD rats were divided into three groups based on their action potential (AP) firing patterns in response to a suprathreshold depolarizing current step (100pA; 5sec). Phasic neurons fired several APs and adapted quickly; tonic‐phasic neurons fired several APs and adapted slowly; tonic neurons fired continuously. After a 1 min exposure to 3mM H 2 O 2 , 20% of tonic neurons were converted to phasic neurons; 80% of tonic neurons continued to fire tonically but at a reduced frequency.
