Apolipoprotein B messenger RNA‐editing, catalytic polypeptide 3 (APOBEC3) family proteins are cellular cytidine deaminases that function as DNA/RNA mutators and play diverse roles in key physiological processes. Recent studies have shown that APOBEC3B, a unique member of this family that predominantly localizes to the nucleus, is implicated in a variety of cancers including squamous cell lung carcinoma, a cancer of which little options are available for targeted therapy. While speculations have been made for a possible association of APOBEC3B to carcinogenesis, little is known regarding the molecular mechanisms that link APOBEC3B to cancer. Biochemical experiments have been conducted to characterize this protein enzymatically, investigating its enzymatic capacity and function, its ability to differentially bind DNA and RNA, and the diverse ways in which APOBEC3B is regulated at the protein level.
