Venous malformations (VMs) are the largest group of vascular malformations of neonates representing more than 50 % of cases. In a large group of spontaneous mutations of VMs the primary cause and the dysregulated genes are unknown. We compared the protein expression in the tunica interna, media and externa of slow-flow VMs and normal saphenous veins with immunofluorescence histochemistry by using selected antibodies. We observed dysregulation in structure, an up-regulation of inflammation markers and "arterialization" of endothelial cells of the VMs. We show that the number of vasa vasorum in the tunica externa and media of the VMs is increased and an up-regulation of inflammation markers CD54, CD62e and CD105 in the vasa vasorum. The structure of the VMs is affected by the number of vasa vasorum and the unstructured morphology of the tunica media. In the endothelium of the VMs we observed up-regulation of the arterial markers EphrinB2 and EphB2 and down-regulation of the venous marker
