葡萄籽原花青素对大鼠脑缺血再灌注后NF-κB、IκB和iNOS的影响

TL;DRAbstract

OBJECTIVE To investigate the grape seed proanthocyamdins extract (GSPE) on the expression of NE-κB, IκB and iNOS in rat after focal cerebral ischemia-reperfusion. METHODS The model of right focal cerebral ischemia reperfusion in rats was established via suture-occluded method. 72 rats were randomly divided, into 3 groups: sham operation group, ischemia-reperfusion and GSPE group. The rats were sacrificed after ischemia for 2 hours and reperfusion for 6, 24 and 48 hours using beheading method. The expression of NE-κB, IκB and iNOS in 1mm tissue and around optic chiasma were examined by immunohistochemical method. RESULTS The expression of NF-κB in ischemia-reperfusion group was significantly higher than GSPE group (P<0.01), while the expression of IκB was significantly lower (P<0.01). CONCLUSION GSPE had protective effects on cerebral ischemia and reperfusion that may relate to prevention IκB from degradation and inhibition of NF-κB activity, down-regutation the expression of iNOS.
