TL;DRAbstract

A major goal of genome research is to isolate the genes and genome loci controlling quantitative traits, i.e., quantitative trait loci (QTL), of scientific and/or economic importance. For the purposes of gene and QTL isolation, several methods have been developed and used. These include: [1] Library screening, [2] DNA subtraction, [3] gene expression differential display including cDNA differential display, microarray or GeneChip, and serial analysis of gene expression (SAGE), [4] expressed sequence tag (EST) and genome sequencing, [5] T-DNA or transposon gene tagging, [6] polymerase chain reaction (PCR)-based candidate gene cloning, [7] antisense- or RNA interference (RNAi)-based gene expression interference, and [8] map-based or positional cloning (hereafter both are referred to as map-based cloning).
