Microencapsulated immunostimating complexes (ISCOMs) as a basis for single-shot vaccines

TL;DRAbstract

To produce a protective immune response, traditional vaccine regimens involve aninitial ‘priming’ dose of antigen followed by repeated booster doses. Thisrequirement is inconvenient for patients (or animals) that need to return so as toreceive the booster doses. The development of single shot vaccines in which apriming dose and booster doses are contained in the same dose form and hencewould only need to be administered once would obviously offer benefits. Further,when a vaccine is based on purified whole or sub-unit antigens, the antigen must bedelivered together with an adjuvant to produce a protective immune response.A particulate system which has received much attention in terms of possibleapplication as a vaccine adjuvant (to the extent that they are now approved for use inveterinary vaccines and are in various stages of human clinical trials) is theimmunostimulating complexes (ISCOMs).
