The isolation and characterization of a rhabdovirus from stone flounder, Kareius bicoloratus.

A strain of rhabdovirus(strain 080113) was isolated from the larvae of cultured stone flounder Kareius bisoloratus.The obvious cytopathic effect(CPE) was observed in 8 cell lines inoculated with tissue homogenates,with the highest titer in Epithelioma papulossum of carp cell line(EPC).The characterization of strain 080113 was studied.The strain was sensitive to temperature,pH3 and pH10,chloroform and not sensitive to 5-iodo-2-deoxyuridine(IUDR),which suggested that the strain was RNA virus with envelope.Typical bullet-shape particles,with the size of(80-118) nm ×(28-55) nm,were observed in the cytoplasm of EPC cells infected by strain 080113.Five protein bands were observed by SDS-PAGE,with the molecular weight of 200 000,56 000,42 000,29 000 and 24 000,respectively.Reverse-transcription polymerase chain reaction(RT-PCR) was tested with the specific primers of infectious hematopoietic necrosis virus(IHNV),viral haemorrhagic septicemia virus(VHSV),hirame rhabdovirus(HRV) and other aquat
