Role of HSV II antigen-specific CD8+ cytotoxic T cells in pathogenesis of patients with genital herpes by pentamer technology.

Objective To investigate the effect of HSV Ⅱ antigen-specific CD8+ cytotoxic T cells(CTLs) in peripheral blood in the pathogensis of the genital herpes.Methods HSV Ⅱ antigen-specific CD8+ cytotoxic T cells from 20 initial and recurrent patients were stained with fluorescent-labeled recombinant MHC class Ⅰ-peptide pentamer complex in peripheral blood.T-test was used to compare the results in two group.Results The percentages of peripheral blood HSV Ⅱ antigen-specific CD8+ cytotoxic T cells in initial patients and recurrent patients were 0.73%±0.29% and 0.35%±0.24%,respectively(P0.001).Conclusions The activity of HSVⅡantigen-specific CD8+ cytotoxic T cells(CTLs) in peripheral blood is enhanced in the former and weakened in the latter.the Pentamer technology is characterized by rapidity,sensitivity,high-efficiency and specificity.
