TL;DRAbstract
The goal of this study was to determine and compare pharmacokinetic parameters of ALT, AST, LD and GD in healthy dogs. Intravenous injection of a liver supernatant was followed by the measurement of the enzymatic activity in plasma during the following 18 days in 7 beagle dogs. The kinetic parameters were determined by a non compartmental analysis. Results of the study: the volumes of distribution in steady-state conditions of ALT, AST, LD and GD were respectively 77 ± 15, 82 ± 51, 51 ± 8 and 57 ± 9 mL/kg, their half-lives were respectively 58.6 ± 8.5, 21.9 ± 14.8, 0.9 ± 0.2 and 28.6 ± 14.5 hours and their plasma clearance were 1.0 ± 0.2, 7.5 ± 1.9, 40.3 ± 11.4 and 2.6 ± 0.4 mL/kg/h respectively. An equation assessing the weight of liver completely destroyed following a period of insult is proposed.
